Noninvasive discrimination of prostate healthy and cancer cells by quantitative phase and Raman imaging

Abstract

In this study, we utilized holotomography to analyze prostate healthy (PNT2) and cancer (PC3) cells treated with glucose. After 48-hour incubation, distinct morphological differences were observed in cancer cells, including changes in lipid droplet volume, number, and refractive index. Raman spectroscopy confirmed the presence of these lipid droplets. Cancer cells exhibited larger and more numerous lipid droplets, with higher mean refractive-index, compared to healthy cells. The study achieved over 90% accuracy in discriminating between cell types, highlighting its potential in cancer diagnostics. The research contributes to biomedical spectroscopy, offering a valuable tool for understanding cancer cell morphology and enabling early and precise cancer diagnosis.

Hossein Khadem earned his Bachelor’s degree in Physics from Ferdowsi University of Mashhad (Iran). He received a Master’s and Ph.D. in Photonics from Shahid Beheshti University, Tehran, Iran. His Master’s project focused on optical properties of gold nanorods for sensing of refractive-index variations in liquids. His doctoral research was on the plasmonic heterodimers, specifically studying the impact of size-asymmetry for surface-enhanced Raman spectroscopy (SERS). Currently, he works as a postdoctoral researcher in the Biophotonics and Advanced Microscopies group, under the guidance of Professor Anna Chiara De Luca, at the IEOS Institute of Experimental Endocrinology and Oncology “G. Salvatore”, National Research Council (CNR), Naples, Italy. His field of research is holotomography and Raman spectroscopy, aiming to label-free detection of cancer cells.
