Integrating light sheet microscopy and multispectral imaging for the analysis of autofluorescent tissues

Abstract

In this work, we present a method for analyzing the autofluorescence signal of an ex-vivo sample, obtained using a custom-made light-sheet microscope system capable of acquiring multispectral images thanks to a tunable filter. Once acquired, Principal Component Analysis (PCA) was employed for spectral unmixing of the captured images since no fluorescent markers are present in the sample.. This tool enables the determination of principal components (PCs) without prior knowledge of spectra. Thus, the extracted PCs can be associated with various specific tissues of the analyzed sample. This method represents a preliminary step toward establishing a non-invasive and label-free tissue characterization approach for both ex-vivo and in-vivo samples.

Dedicated to research and teaching since 2013, I finished my Ph.D. in Physics Applied to Sciences and Technologies at the University of Alicante in 2017. Currently holding an assistant professor position at the University of Alicante, my research focuses on optics, holography, and biomedical imaging. In optics and holography, I work in the field of the photopolymers, specifically in the recording of complex diffractive optical elements, and the recording of holographic waveguides in these materials, applicable to photovoltaic energy and displays. In biomedical imaging, my work involves various imaging systems for tissue characterization and invertebrate behavior analysis as well and early tumor detection systems. As an educator, I teach courses in telecommunications engineering, robotics engineering, and biomedical engineering, emphasizing signal processing and biomedical imaging courses.
